Like most analytical instruments, liquid chromatographs have certain requirements for the environment around the instrument, but there are no special requirements to meet the basic conditions of the general analytical instrument room, which can ensure the stable operation and safe operation of the instrument.

(1) Lighting:

Indoor lighting should be light and avoid direct sunlight. To avoid the effects of direct light on the operation and observation of the instrument, the instrument (display) should be laterally illuminating. Window curtains should be hung in the laboratory window to adjust the light intensity at any time.

(2) Ventilation:

During the analysis using liquid chromatograph, since the mobile phase contains volatile organic solvents (methanol, acetonitrile, etc.), harmful gases are generated, causing corrosion to the optical components in the instrument. Attention should be paid to the ventilation of the instrument room to eliminate indoors. Harmful gases and keep the air fresh.

There are two main ways of ventilation: one is natural ventilation, the doors and windows are opened regularly to allow convection of the inside and outside air, and the other is forced ventilation, such as using a fume hood, exhaust fan, etc., to discharge harmful gases out of the room in time.

(3) Ambient temperature and humidity:

Appropriate temperature and humidity are necessary to ensure the normal operation and stable performance of the HPLC instrument.

The HPLC instrument generally requires a room temperature between 20 and 30 ° C, and the daily temperature changes by 2 to 3 ° C. Do not fluctuate too much. Otherwise, the chromatographic separation will be affected during the startup analysis;

In the summer, it is necessary to control the room temperature not to exceed 30 ° C. If the temperature is too high, bubbles will be generated in the flow system, which will affect the normal operation of the detector (such as fluorescence detector). The indoor relative humidity should be controlled below 60%.

(4) Power supply:

The liquid chromatography room should be provided with a lighting power source and a power source, and the power source can be used for the operation of the instrument. In order to reduce noise interference, the instrument should be away from high-power electrical equipment that is instantaneously powered, such as large electric equipment (ultra-low temperature refrigerator, electric oven, etc.), elevators, air conditioning systems, etc. In order to ensure the accuracy of the HPLC infusion pump, it is necessary to ensure a stable input voltage. For this reason, it is recommended that the chromatographic chamber be provided with a regulated power supply equivalent to the instrument load. The grounding wire should be installed on the indoor wall to meet the requirements of the grounding wire. The common grounding resistance of the instrument for protective grounding is ≤4Ω.

(5) Cleanliness:

The particulate matter suspended in the air will interfere with the analysis of the chromatographic instrument, increase the background and noise, and become a pollution source for analytical testing, directly affecting the purity of high-purity materials and the analysis of trace impurities.

The HPLC instrument is a benchtop, the detector and the optical path system are enclosed inside the instrument. The interference from the ground and air is small. Generally, no buffer isolation is required, but care should be taken to keep the room clean, especially when the instrument is placed on one floor. Because the dust in the air is large, in order to intercept the source of particulate matter pollution, the ground and the table top should be cleaned frequently so that the instrument does not accumulate dust or adsorb dust.

Especially before starting the machine, the environment around the instrument should be cleaned to prevent the surrounding dust from being absorbed after the instrument is energized, which will cause interference to the analysis.
